Study sites

We have now collected samples at two sites in Pennsylvanian oak forests, one site in Indiana, and one site in Maryland. The first site is near the Powdermill Biological Station 60 miles southeast of Pittsburgh and the second is in the Hawk Mountain Sanctuary about 50 miles southwest of Wilkes-Barre. Sites outside of Pennsylvania include Savage River, a state park of Maryland, and The Pioneer Mothers Memorial Forest, in the Hoosier National Forest of Indiana. At both sites we set up an 8 by 100 meter transect. Within this transect we identified, mapped the location of and collected leaf samples from every oak seedling. We also identified, mapped the location of and collected leaf samples from every adult oak tree within 30 meters of the transect (i.e. in a 60 by 160 meter grid).

We then extracted DNA from every leaf sample and determined its DNA fingerpint by performing PCR with 10 different primer sets which amplify polymorphic microsatellite loci.
